Synthetic Approaches towards Peptidomimetic Design

چکیده
• Modified peptides: these molecules contain relatively small modifications that do not modify the peptide bond, and thus they still possess a chemical structure of peptide nature. • Pseudopeptides: in these compounds partial modifications of either peptide bonds or side-chains are introduced, contributing to the generation of molecules possessing a chemical structure of only partial peptide nature. • Peptidomimetics: these compounds do not possess any amide bond of the original peptide, and the structural resemblance is related to the pharmacophore and to the pharmacological activity of the bioactive peptide conformation.

Protein ligand design: from phage display to synthetic protein epitope mimetics in human antibody Fc-binding peptidomimetics.
Phage display is a powerful method for selecting peptides with novel binding functions. Synthetic peptidomimetic chemistry is a powerful tool for creating structural diversity in ligands as a means to establish structure-activity relationships. Here we illustrate a method of bridging these two methodologies, by starting with a disulfide bridged phage display peptide which binds a human antibody...
متن کاملAdaptive Evolution of Escherichia coli to an α-Peptide/β-Peptoid Peptidomimetic Induces Stable Resistance
Antimicrobial peptides (AMPs) and synthetic analogues thereof target conserved structures of bacterial cell envelopes and hence, development of resistance has been considered an unlikely event. However, recently bacterial resistance to AMPs has been observed, and the aim of the present study was to determine whether bacterial resistance may also evolve against synthetic AMP analogues, e.g. α-pe...
متن کاملProtease-resistant peptide design-empowering nature's fragile warriors against HIV.
Peptides have great potential as therapeutic agents, but their use is often limited by susceptibility to proteolysis and their resulting in vivo fragility. In this review, we focus on peptidomimetic approaches to produce protease-resistant peptides with the potential for greatly improved clinical utility. We focus on the use of mirror-image (D-peptide) and ß-peptides as two leading approaches w...
متن کاملSynthesis and conformational studies of a stable peptidomimetic -hairpin based on a bifunctional diketopiperazine turn inducer
The design, synthesis and conformational studies of a new b-hairpin mimic, 2, are described in this paper. The design towards hairpin 2 is based on the assembly of the bifunctional DKP-1 as a b-turn inducer, a peptidomimetic strand, namely 5-amino-2-methoxybenzhydrazide, to stabilize the formation of b-sheets, and finally a tetrapeptide sequence, GVVI, containing the hydrophobic residues Val, I...
متن کاملAn Adaptive Geometric Search Algorithm for Macromolecular Scaffold Selection
15 A wide variety of protein and peptidomimetic design tasks require matching 16 functional three-dimensional motifs to potential oligomeric scaffolds. Enzyme 17 design, for example, aims to graft active-site patterns typically consisting of 3 to 18 15 residues onto new protein surfaces. Identifying suitable proteins capable of 19 scaffolding such active-site engraftment requires costly searche...
